I have tested the game without the SAFEMODE=1 added to my launch properties after running it on first launch and it starts and runs just fine. I went through the first floor with no issues.
I think safe mode is needed to forcibly place the settings file in the proper directory so you can edit it from there.
I haven't tested full screen yet but I will when I get back from work.
