Recommend fall back to stable Wine 1.2 series. Running Vocaloid2 under Wine 1.3 is very buggy. Works cannot be done.

HowTo / Notes

Using under Jackd

Set Vocaloid2 output to Pulseaudio in Vocaloid2 editor. Then use pulseaudio-jack bridge,to send into  Jackd.

It work perfectly. I have also tested directly output into jack. As I remembered, it didn't work well.
