Everything works great! but info about upgrades to buildings: I think an alt-text or pop-up just doesn't appear.
What does not
fullscreen: you must emulate virtual desktop and set to 1280-1024.
And referrals to disk path don't work unless you insert the physical disk.
maybe it needs an .iso file to refer to; but I haven't figured out how to make one on a mac :)
What was not tested
Lovely game, great game engine, lots of jokes and challenging levels.

Nothing special to mind, simply run
$ WINEPREFIX=/path/to/Tropico/prefix wine /path/to/_setup/Setup.exe
You should cd into the installation directory, otherwise Tropico might fail with an empty error box. Therefore the invocation looks like (if you installed into the default directory):
$ cd /path/to/Tropico/prefix/drive_c/Program\ files/Troico && WINEPREFIX=/path/to/Tropico/prefix wine Tropico.EXE
No other settings are needed to become El Presidente.
