What works
The game loads and runs. Graphics and sound all look good.
What does not
The game does not detect the RealTone USB cable used to connect the guitar to the computer. The cable is a simple USB audio device, but apparently they take some anti-piracy measures to force users to use an official cable. It's the same cable as was used in the original RockSmith, and I believe players had to use a "no cable crack" to run that program.
Workarounds
What was not tested
Actual game-play was not tested, since it's not possible to get to that point without the game recognizing the cable. However, there's every indication that it would work if not for that one hangup.

DirectX/Direct3D is necessary for it to run. I used winetricks to install: d3dx9 d3dx10 d3dx11_43 corefonts vcrun2005 vcrun2008 vcrun2010 msxml4 xact although I'm not sure that all of those were necessary. Additionally, it was necessary to set a couple things in the Rocksmith.ini file: ForceWDM=1 (to avoid a page fault) Win32UltraLowLatencyMode=0 (to make the audio sound good)
