What works
Everything works, once you get the audio configuration right.
Setup WINE to Vista, Windows 7 or newer.
Install under Steam, no problems.
Set
Win32UltraLowLatencyMode=0
in Rocksmith.ini
There are at least 2 configurations that work (on a distro with pulseaudio):
1. Use pulseaudio for everything. This is bad because of the audio latency. Also your guitar cable detection is somewhat random and the capture crackles quite easily with increasing CPU load, causing note detection to fail.
2. Use wine ALSA driver for audio and pulseaudio driver for ALSA output. Using HW output directly did not work, since starting Steam blocks the audio device, independent of the "Audio exclusivity" setting. (This a wine bug? It should give rocksmith all the audio devices with that enabled?). I did not have to switch off the Rocksmith cable from pulseaudio manager.
There was no problems with the steam overlay either. Audio latency was ok, so it's playable without a separate guitar amp.
What does not
It has various visual bugs with less mature graphics drivers (nouveau+GT920M). No crashes though. With nvidia binary driver on a GTS450 visuals are flawless. Needs a lot of CPU power so your sound does not crackle (pulseaudio).
Workarounds
What was not tested
Guitararcade was not tested, apart from Score Attack, which works flawlessly. Also wineasio + jack configuration was not tested since ALSA/PulseAudio worked well enough.

Additional Comments
Audio exclusivity in Wine does not seem to work. Once Steam is started no application has access to the output device, except with ALSA pulseaudio driver. It would be handy to have the option to select the Wine audio driver from the Wine configuration app back, with automatic selection this game would not work in any circumstance. The Rocksmith cable was shown on the audio device list very randomly . (I used Audacity under Wine to test this) With Wine forced to use ALSA driver it is stable. Tested systems: Desktop with i5-2500k CPU, Nvidia GTS 450 and Realtek HD Audio, Fedora 23 64-bit kernel 4.4.5 with nvidia binary driver. Laptop with i5-5200U, Nvidia GT920M/Intel HD4400 and Intel HD Audio, Fedora 23 64-bit kernel 4.4.5 with nouveau driver.
