What works
The game itself works flawlessly, no hangups, tools ok, etcetera. I tried it using a zip file which already had all the files there (no installation). There are a few things that do not work which i will comment on below, but anyway, the game is perfectly playable.
Loading/Saving works fine, except for the first save (see point 4 in "does not work")
What does not
Problems I've found:
1) It may ask for some DLL and INI file, though just clicking OK is enough. Alternatively, you may copy those files to the required folders.
2) After this, it said some error about no soundcard detected, and it asks whether you want to run the game or not. Clicking yes loads the game without trouble. However, as you may have guessed, you won't have sound (you are not missing much anyway...)
3) You can't increase the size of the window or play fullscreen (it will just show blurry shit in the expanded area). You can, however, make the screen smaller. I am however using 1280*1024, at a lower resolution this may not be an issue.
4) There is a probling saving new games, once you have a saved tower it is no problem to save it again, but to start saving you'll need to make a .TDT file (savegame file) and save over it (it will ask for overwriting). Besides the "save as..." dialog shows a list of files and an "open" command (though the problem is in the caption, the button works fine). Creating a blank file and renaming it as .tdt for overwriting works.
Bottom line: The only thing that does not work is sound, though you are not missing that much.

At present, do NOT attempt to play SimTower with sound support enabled.
It will crash or hang infinitely, depending on your Windows version setting. In either case, it's not playable.
Disable sound, either by editing SIMTOWER.INI or disabling your sound device in winecfg's audio tab.
This is true as of 1.1.13 and today's latest git (1-23-09).
